When Fragment Enrichment Makes More Sense Than Standard cfDNA Extraction

Not every plasma cfDNA workflow is really trying to recover as much total DNA as possible. In some cases, what matters more is whether the sample better reflects the short-fragment population that is analytically relevant downstream.

That is where fragment enrichment begins to make sense. Instead of recovering total cfDNA directly, enrichment workflows introduce a size-selection stage before final capture. This makes the route different in principle from standard extraction.

This distinction matters because fragment profile is not always just a side effect. In liquid biopsy and short-fragment-biased applications, representation can matter as much as raw recovery. A workflow that brings back a broad DNA population may still be less useful than one that reduces large-fragment background and improves the relative signal of shorter cfDNA fractions.

Of course, enrichment is not the answer to every cfDNA problem. Stronger size selection may improve short-fragment representation, but it can also reduce recovery of longer cfDNA fractions. It is most meaningful when fragment behavior itself becomes part of the analytical objective.
